How Semax Works

Semax is a synthetic heptapeptide derived from the ACTH(4-10) fragment (a portion of adrenocorticotropic hormone). Unlike ACTH itself, Semax has no hormonal activity on the adrenal glands — the relevant portion is the fragment that modulates brain activity.

Its primary characterised mechanism is BDNF upregulation: Semax significantly increases the expression of BDNF and its receptor TrkB in the hippocampus — a region central to memory, learning, and mood regulation. BDNF supports neuronal survival, synaptic plasticity, and the formation of new connections.

Semax also influences:

  • Dopamine and serotonin turnover in several brain regions
  • VEGF (vascular endothelial growth factor) — relevant to cerebral blood flow
  • Neuroprotective mechanisms — studied in the context of stroke recovery and neurodegeneration in animal models

The user experience of Semax in research contexts is often described as mentally energising — improved working memory, sharper attention, and faster processing. At higher doses, some researchers note slight anxiety or overstimulation.

How Selank Works

Selank is a synthetic analogue of tuftsin — a naturally occurring tetrapeptide with immune-modulating properties. The synthetic version adds a stabilising sequence that extends its half-life.

Its primary mechanism involves the GABA-A receptor system and serotonin pathways:

  • Modulates GABA-A receptor sensitivity, producing anxiolytic effects
  • Increases serotonin turnover in certain brain regions
  • Shows enkephalin-like (opioid receptor) activity in some studies — possibly contributing to its mood-stabilising profile

Selank's anxiolytic effect is notably different from benzodiazepines — it does not cause sedation or cognitive impairment at standard research doses, and no dependence or withdrawal has been documented in animal studies. This profile is what distinguishes it from conventional anxiolytics.

Immunomodulatory effects have also been documented — Selank appears to influence immune cell activity, though the clinical significance in healthy subjects is unclear.

Semax is studied for:

Evidence Context

Both compounds were developed and studied primarily within the Russian research system. While they are approved medications in Russia for cognitive and anxiolytic indications respectively, their evidence base in Western peer-reviewed literature is limited. Most publications are from Russian institutions, and large randomised controlled trials meeting current Western standards have not been conducted.

This does not invalidate the research — but it means extrapolation to a broader population requires appropriate caution.
