The triple-agonist mechanism means retatrutide works across appetite suppression, insulin secretion, and energy expenditure at the same time. Phase II clinical trial data published by Jastreboff et al. (2023) in the New England Journal of Medicine showed mean body weight reduction of up to 24.2% over 48 weeks — the largest reduction recorded in any incretin-class peptide trial.
It remains an investigational compound. Not approved for medical use by the FDA or any equivalent authority.

Phase II trial (Jastreboff et al., 2023): 338 participants, 48 weeks, mean weight loss up to 24.2% at the highest dose. Significant reductions in waist circumference, blood pressure, and lipid markers also recorded.
Mechanism: GLP-1 suppresses appetite and slows gastric emptying. GIP enhances insulin response and fat metabolism. Glucagon receptor activation increases energy expenditure — the component that distinguishes retatrutide from tirzepatide.
Side effects in trials: Nausea, vomiting, decreased appetite — consistent with GLP-1 class, mild to moderate, dose-dependent.
