How GHK-Cu Works

GHK-Cu (Glycyl-L-Histidyl-L-Lysine Copper) is a naturally occurring copper-binding tripeptide found in human plasma, urine, and saliva. Its plasma concentration declines with age — from approximately 200 ng/mL at age 20 to less than 80 ng/mL by age 60 — tracking closely with the age-related decline in wound healing and skin quality.

GHK-Cu acts as a biological switch for tissue repair and remodelling:

  • Upregulates collagen and glycosaminoglycan synthesis
  • Activates matrix metalloproteinases (MMPs) for collagen remodelling
  • Inhibits TGF-β1 — which reduces excessive scar formation
  • Stimulates angiogenesis (new blood vessel formation)
  • Activates over 4,000 genes according to genome-wide analysis (Pickart & Margolina, 2018), with effects spanning wound healing, anti-inflammatory, and antioxidant pathways

Topically, GHK-Cu has been studied in clinical trials showing improvements in skin thickness, elasticity, and wrinkle depth. Injectable GHK-Cu is studied for systemic and deeper tissue effects.

How Collagen Supplements Work

Hydrolysed collagen (collagen peptides) is collagen protein that has been broken down into smaller peptides and amino acids, making it absorbable orally. These fragments are digested, absorbed, and used by the body as:

  1. Amino acid substrates: Glycine, proline, and hydroxyproline are required in high quantities for collagen synthesis. Increasing their availability supports fibroblast productivity.

  2. Bioactive peptide signals: Some small collagen peptides (particularly dipeptides and tripeptides containing hydroxyproline) appear to stimulate fibroblast collagen synthesis when absorbed intact. This has been demonstrated in cell culture and some human skin trials.

Multiple systematic reviews have found collagen supplements improve skin elasticity, hydration, and wrinkle depth in placebo-controlled trials — with the strongest evidence in older populations where baseline collagen production is lower.

Which Is More Useful for What

For skin quality and anti-aging (topical application): GHK-Cu has direct evidence at the site of application — topical formulations with 2–3% GHK-Cu have shown measurable improvements in skin thickness and fine lines in clinical studies.

For systemic connective tissue support (joints, tendons, gut): Collagen supplements have more robust evidence here. The hydrolysed format reaches systemic tissues and provides the amino acid substrate where it is needed.

For activating deeper repair processes (injectable research): Injectable GHK-Cu targets the signalling pathway — turning on repair biology in a way that oral collagen cannot.

They address the same broad goal (collagen health) through entirely different mechanisms. Neither replaces the other.
