What is MOTS-c?
MOTS-c is a mitochondrial-derived peptide — encoded within mitochondrial DNA rather than nuclear DNA, making it unusual among researched peptides. First identified by Lee et al. in 2015, it has become one of the most studied peptides in longevity and metabolic research.
It regulates metabolic homeostasis by acting as a mitochondrial signal that communicates energy status to the cell and triggers adaptive responses to metabolic stress. MOTS-c levels decline with age and researchers have proposed it as a key mediator of exercise-related metabolic benefits.
What the research shows
Metabolic regulation: Mouse studies show improved insulin sensitivity, reduced fat accumulation, and prevention of diet-induced obesity — even without changes in food intake.
Exercise mimicry: Activates AMPK and downstream pathways also activated by exercise, leading to descriptions of it as an exercise mimetic.
Aging: MOTS-c levels decline with age in animal and human studies. Higher circulating levels correlate with better metabolic profiles.
Muscle: Supports muscle function and endurance in aged animal models.
