What is IGF-1 LR3?
Long R3 IGF-1, commonly written as IGF-1 LR3, is an engineered analogue of insulin-like growth factor 1. It contains an N-terminal extension and an arginine substitution at position three of the IGF-1 sequence.
These structural changes were developed as research tools to reduce affinity for insulin-like growth factor binding proteins while retaining activity at the IGF-1 receptor. This allows researchers to study IGF signalling under conditions that differ from native IGF-1.
Published IGF-1 LR3 research is predominantly preclinical, including cell and animal models. ASCEND is an investigational research product and is not approved for medical use.
What the research shows
The evidence base for IGF-1 LR3 is not comparable with large human clinical programmes. The most relevant findings concern its design and use as a laboratory analogue:
Analogue design: The Long R3 structure combines an N-terminal extension with replacement of glutamate by arginine at position three of the IGF-1 sequence.
Binding-protein affinity: Early analogue studies found markedly reduced interaction with IGF binding proteins compared with native IGF-1.
Cell-model potency: In cell lines that secrete IGF binding proteins, Long R3 IGF-1 showed greater apparent biological potency than native IGF-1, supporting its use as a research reagent.
Evidence limits: Cell and animal findings cannot establish safety, efficacy, or dosing for people, and should not be presented as clinical outcomes.
